چکیده
The southern white rhinoceros (Ceratotherium simum simum) faces an uncertain future in the wild due to increased poaching pressure and habitat fragmentation, thus management of reproductively successful populations is critical importance. Successful reproductive outcomes may be mediated by diet gut microbial diversity; therefore, understanding dynamics within between captive help improve conservation efforts. Accordingly, microbiome preservation methods are needed that practical for situ field sampling populations. We evaluated efficacy 3 different over 2 timepoints stabilizing communities feces from (n = 10) at North Carolina Zoo Asheboro, Carolina, USA, during July–September 2020 January–March 2021. Samples were immediately frozen −80°C, stored PERFORMAbiome™·GUT (PB) tubes or 95% ethanol ambient temperatures (to simulate conditions), processed after 14 230 days post-collection. quantitatively compared alpha beta diversity across identified taxa enriched each treatment group. preserved consistently harbored lower Shannon index (SHDI) Simpson's (SDI) values Frozen PB samples. This trend was apparent both Ethanol day-14 samples (SHDI 4.94; SDI 0.98) versus 5.19; W 518, P < 0.001; 0.99; 476, 0.001) 5.15; 430, 0.01; 1075, 1) samples, day-230 4.48; 0.97) 5.18; 0, 0.05; 0.032) 5.23; day differed (P 0.05) all other treatments indices. Notably, maintained compositionally similar time points. Our results indicate perform similarly freezing highlighting their utility fieldwork applications. Identifying optimal versatile techniques will enable studies reproductively-successful populations, effort central efforts threatened species.

Fecal corticosterone concentrations and reproductive success in captive female southern white rhinoceros.
Prolonged or frequent secretion of adrenal glucocorticoids in response to aversive stimuli can negatively impact reproduction. Because female southern white rhinoceros (Ceratotherium simum simum) reproduce poorly in captivity, we compared fecal corticosterone metabolite concentrations among parous, nulliparous, and adolescent females and examined social and physical aspects of the captive envir...
متن کاملSocial and spatial relationships in captive southern white rhinoceros (Ceratotherium simum simum).
Although critical to the conservation of white rhinoceros, captive breeding has proven challenging because of the poor and irregular reproductive health of many captive rhinos, and social interactions may play a significant role. This research investigated the social and spatial relationships of two captive groups of southern white rhinoceros (Ceratotherium simum simum) by examining the frequen...
متن کاملControl of the gut microbiome by fecal microRNA
Since their discovery in the early 90s, microRNAs (miRNAs), small non-coding RNAs, have mainly been associated with posttranscriptional regulation of gene expression on a cell-autonomous level. Recent evidence has extended this role by adding inter-species communication to the manifold functional range. In our latest study [Liu S, et al., 2016, Cell Host & Microbe], we identified miRNAs in gut ...
متن کاملRecovery of the Gut Microbiome following Fecal Microbiota Transplantation
UNLABELLED Clostridium difficile infection is one of the most common health care-associated infections, and up to 40% of patients suffer from recurrence of disease following standard antibiotic therapy. Recently, fecal microbiota transplantation (FMT) has been successfully used to treat recurrent C. difficile infection. It is hypothesized that FMT aids in recovery of a microbiota capable of col...
متن کاملPreservation of three-dimensional spatial structure in the gut microbiome
Preservation of three-dimensional structure in the gut is necessary in order to analyze the spatial organization of the gut microbiota and gut luminal contents. In this study, we evaluated preparation methods for mouse gut with the goal of preserving micron-scale spatial structure while performing fluorescence imaging assays. Our evaluation of embedding methods showed that commonly used media s...
